Ang-2 over-expression impairs islet function but protects from cytokine treatment in isolated islets.
Isolated islets from RIP-rtTA;tet-O-Ang-2 (Ang2-rtTA) and RIP-rtTA control (rtTA) mice were cultured for 3 days in presence of 10 μg/ml doxycycline to achieve Ang-2 overexpression. Mouse or human islets were cultured in 11.1 (mouse) or 5.5 mM glucose (human) or treated with diabetic conditions of 22.2 mM glucose + 0.5mM palmitic acid or mixture of cytokines: 2 ng/mL IL-1β, 1000 U/ml IFN-ɣ and TNF-α (cyto). Ang pag-aaral ay papatungkol sa buhay, pag-aakda, at kultural na produksiyon ng itinuturing na Ama ng Kontemporanyong Panitikan ng Kanlurang Visayas na si Leoncio P. Deriada. Tinugunan ng pag-aaral ang hamon ng kritiko at pambansang alagad ng sining na si Bienvenido Lumbera na lumikha ng talambuhay ng mga alagad ng sining ng rehiyon upang mapunan ang puwang sa kung paano idurugtong at ipagsasanib ang panitikan ng rehiyon at ang itinuturing na panitikan ng bansa. Kung kaya’t nilayon ng pag-aaral na itampok ang buhay at pag-aakda ni Leoncio P. Deriada bilang sagot sa hamon at pangarap na ito sa pagtatanghal ng talambuhay ng naturang manlilikha na bukod sa layunin ng dokumentasyon ay nagsulong din ng rehiyonal na panitikan sa kamalayan ng mas maraming mamamayan na ang mas nakilala lamang na panitikang pambansa ay yaong mga naunang isinubo ng sistema ng edukasyon na madalas ay mga akdang mas nagmumula sa sentro o sa Maynila . Ililitaw ng pag-aaral ang mahalagang papel ni Deriada upang mamayagpag ang kontemporanyong panitikan ng Kanlurang Visayas lalo pa ang pagpapakilala ng mga wikang Hiligaynon, Kinaray-a, at Aklanon, bukod sa Filipino at Ingles, bilang mga wika ng pambansang panitikan. Sa kanyang naging paglalandas upang maging ganap ang kanyang persona bilang kabataan, estudyante, asawa’t ama, manunulat at manggagawang-pangkultura, nabuo ang mga antolohiya ng mga rehiyonal at pambansang manunulat at akda bilang kongkretisasyon ng konsepto niya ng panitikan ng rehiyon na sumuhay sa panitikan ng bansa. Kung paanong ang pag-aaral sa buhay at pag-aakda ni Deriada ay nagpatibay din sa argumento na ang panitikan ng bansa ay marami, hindi iisa, walang sentro, dahil lahat ng rehiyon ay sentro ng panitikang pambansa. Tinalakay sa tesis na ito ang tunggalian ng mga kasariang matatagpuan sa nobela nina Alejandro G. Abadilla at Elpidio P. Kapulong na Ang Pagkamulat ni Magdalena. Upang magawa ito, hinalaw ko ang ilang konsepto ni Kate Millett sa kanyang teorya ng Pulitikang Pangkasarian partikular na ang naging diskusyon niya sa mga akda ni Norman Mailer. Kasabay nito ginamit ko rin ang konsepto ng Maria Magdalena sa Biblia at iniangkop ito sa buhay ng pangunahing tauhan ng nobela na si Denang (Palayaw ni Magdalena) na maihahalintulad sa kanya. Angiotensin II induces soluble fms-Like tyrosine kinase-1 release via calcineurin signaling pathway in pregnancy
Maternal endothelial dysfunction in preeclampsia is associated with increased soluble fms-like tyrosine kinase-1 (sFlt-1), a circulating antagonist of vascular endothelial growth factor and placental growth factor. Angiotensin II (Ang II) is a potent vasoconstrictor that increases concomitant with sFlt-1 during pregnancy. Therefore, we speculated that Ang II may promote the expression of sFlt-1 in pregnancy. Here we report that infusion of Ang II significantly increases circulating levels of sFlt-1 in pregnant mice, thereby demonstrating that Ang II is a regulator of sFlt-1 secretion in vivo. Furthermore, Ang II stimulated sFlt-1 production in a dose- and time-dependent manner from human villous explants and cultured trophoblasts but not from endothelial cells, suggesting that trophoblasts are the primary source of sFlt-1 during pregnancy. As expected, Ang II-induced sFlt-1 secretion resulted in the inhibition of endothelial cell migration and in vitro tube formation. In vitro and in vivo studies with losartan, small interfering RNA specific for calcineurin and FK506 demonstrated that Ang II-mediated sFlt-1 release was via Ang II type 1 receptor activation and calcineurin signaling, respectively. These findings reveal a previously unrecognized regulatory role for Ang II on sFlt-1 expression in murine and human pregnancy and suggest that elevated sFlt-1 levels in preeclampsia may be caused by a dysregulation of the local renin/angiotensin system

Sa kasalukuyang yugto ng hyphenated identity isinilang ang HORI7ON (호라이즌), ang kauna-unahang Filipino boy band na nagdebut sa South Korea noong 24 Hulyo 2023. Kinabibilangan nina Jeromy Batac, Marcus Cabais, Kyler Chua, Vinci Malizon, Reyster Yton, Kim Ng, at Winston Pineda ang grupo. Sila ang pitong nanguna sa animnapu’t dalawang kalahok ng survival competition series na Dream Maker na inere sa A2Z at Kapamilya Channel—sa cable at satellite TV, Facebook, at YouTube—mula 19 Nobyembre 2022 hanggang 12 Pebrero 2023. Nang pirmahan ng ABS-CBN at ng mga South Korean na entidad na MLD Entertainment at KAMP Global ang kontrata sa paglulunsad ng Dream Maker, G-pop o Global Pop ang marketing sa mabubuong grupo, at hindi isang K-pop o Korean Pop group. Gayunman, kinukutya ng ilang fans ng P-pop o Pinoy Pop ang HORI7ON, sapagkat hindi diumano isang tunay na P-pop group ang HORI7ON, at ni hindi sila dapat nagtanghal sa Pinoy Pop Convention and Concert (PPOPCON) noong Hulyo 2023. Sisikapin ng papel na ito na suriin ang pagbuo sa at pag-iral ng HORI7ON bilang kadugtong ng Hallyu o Korean Wave sa postkolonyal na kalagayan ng Pilipinas. Representative low-magnification light photomicrographs display H&E staining (Fig 6(a)), Masson's Trichrome staining (Fig 6(b)) and PAS stain Fig 6(c)) of whole lung samples from PBS-challenged/ vehicle treated (n = 6) (PBS), OVA-challenged/ vehicle treated (n = 6) (OVA), OVA-challenged/Ang-(1–7) treated (0.3 mg/kg; n = 6) (Ang-1–7), OVA-challenged/Ang-(1–7) and A779 treated (0.3 mg/kg; n = 6 and 1mg/kg; n = 5; respectively) (Ang-1–7 + A779), OVA-challenged/dexamethasone treated (1 mg/kg; n = 6) (DEX). OVA-challenged/vehicle treated mice showed marked and significant peribronchial and perivascular inflammatory cell infiltrations (a) peribronchial and perivascular fibrosis (b) and bronchial mucus production and goblet cell hyper/metaplasia (c) compared with PBS-challenged vehicle treated mice. Treatment with Ang-(1–7) resulted in a significant reduction in the peribronchial and perivascular dark-staining inflammatory cell infiltration (a), peribronchial and perivascular fibrosis (b) and bronchial mucus production and goblet cell hyper/metaplasia (c) compared to the OVA – challenged mice and was comparable to PBS-challenged and OVA-challenged/dexamethasone treated mice.
