    Proline-rich tyrosine kinase 2 mediates gonadotropin-releasing hormone signaling to a specific extracellularly regulated kinase-sensitive transcriptional locus in the luteinizing hormone beta-subunit gene

    G protein-coupled receptor regulation of gene transcription primarily occurs through the phosphorylation of transcription factors by MAPKs. This requires transduction of an activating signal via scaffold proteins that can ultimately determine the outcome by binding signaling kinases and adapter proteins with effects on the target transcription factor and locus of activation. By investigating these mechanisms, we have elucidated how pituitary gonadotrope cells decode an input GnRH signal into coherent transcriptional output from the LH beta-subunit gene promoter. We show that GnRH activates c-Src and multiple members of the MAPK family, c-Jun NH2-terminal kinase 1/2, p38MAPK, and ERK1/2. Using dominant-negative point mutations and chemical inhibitors, we identified that calcium-dependent proline-rich tyrosine kinase 2 specifically acts as a scaffold for a focal adhesion/cytoskeleton-dependent complex comprised of c-Src, Grb2, and mSos that translocates an ERK-activating signal to the nucleus. The locus of action of ERK was specifically mapped to early growth response-1 (Egr-1) DNA binding sites within the LH beta-subunit gene proximal promoter, which was also activated by p38MAPK, but not c-Jun NH2-terminal kinase 1/2. Egr-1 was confirmed as the transcription factor target of ERK and p38MAPK by blockade of protein expression, transcriptional activity, and DNA binding. We have identified a novel GnRH-activated proline-rich tyrosine kinase 2-dependent ERK-mediated signal transduction pathway that specifically regulates Egr-1 activation of the LH beta-subunit proximal gene promoter, and thus provide insight into the molecular mechanisms required for differential regulation of gonadotropin gene expression

Considering f(0), we give more sharp estimations for them where nonnegativity of f is not needed.     Calculation of Three-dimensional Unsteady Sheet Cavitation by a Simple Surface Panel Method "SQCM"

    This paper presents a calculation method for the 3-D unsteady cavitating hydrofoil problem. The method is based on a simple surface panel method "SQCM" which satisfies easily the Kutta condition even in the unsteady problem. This method is applied to the Wagner problem, heaving hydrofoil and the hydrofoil in sinusoidal gust. We show some calculated results for partially cavitating and supercavitating hydrofoil and compare them with other calculated results

    A significant example in the history of the stereotomy is provided by a special ‘barrel-vault’ stone device which created roofing for spiral staircases. Already known and used in the twelfth century, this surface was first mentioned in a treatise at the end of the fourth book of Le premier tome de l’Architecture by Philibert De l’Orme: among the various examples of spiral staircase there is the first reference to a spiral barrel vault as the Saint-Gilles screw or spiral staircase. The name of this surface derives from the presence of an example of vault roofing whose intrados is characterised by the above-mentioned surface at the Abbey of Saint-Gilles in France. Although the staircase was completed in 1142, long before the first rigorous formulations of stone-cutting, the excellence and accuracy of the construction ensured that the ‘vis de Saint-Gilles’ became consecrated as a vital stage of the pilgrimages made by stone masons’ corporations to visit the works of greatest importance for learning the art of stereotomy. Evidence of their visits is provided by the marks engraved in the side walls of the staircase left by members of the corporations

    This paper provides an empirical analysis of small and medium enterprise (SME) participation in production networks. It gauges firm characteristic determinants of SME participation in production networks. The empirical investigation utilizes results obtained from an ERIA Survey on SME Participation in Production Networks, conducted over a three month period at the end 2009 in most ASEAN countries (i.e., Thailand, Indonesia, Malaysia, Philippines, Vietnam, Cambodia, and Laos PDR) and China. The results suggest that productivity, foreign ownership, financial characteristics, innovation efforts, and managerial/entrepreneurial attitudes are the important firm characteristics that determine SME participation in production networks. The paper extends the analysis to identify the determinants that allow SMEs to move from low to high quality or value adding participation in production networks. The results suggest that size, productivity, foreign ownership, and, to some extent, innovation efforts and managerial attitudes, are the important firm characteristics needed by SMEs to upgrade their positions in production networks. The finding suggests that SMEs really exploit competitiveness from economies of scale only when they are able to engage in production networks.
