Kinetics of the lipid metabolism in a mouse model of the overexpression of the lysosomal acid lipase
Die Lysosomale Saure Lipase spielt eine zentrale Rolle im Stoffwechsel der Lipide. Sie katalysiert die intrazelluläre Hydrolyse von Triacylglycerolen und Cholesterolestern in den Lysosomen, wobei Cholesterol und freie Fettsäuren entstehen. Ein Mangel dieses Enzyms führt zur Akkumulation von Cholesterolestern und Triacylglycerolen in den Zellen. Die Wolman Krankheit und Cholesterolester Speichererkrankung (CESD) sind zwei seltene Erkrankungen mit einem angeborenen Mangel dieses Enzymes. Die pathophysiologischen Vorgänge des dabei vorliegenden Stoffwechsels konnten in Tiermodellen bereits nachvollzogen werden.
Zur weiteren Untersuchung wurde von einer Forschergruppe ein Tiermodell entwickelt, das die Lysosomale Saure Lipase über exprimiert. Bei dessen Charakterisierung wurde eine erhöhte Lipi-deinspeicherung in der Leber, sowie einer erhöhte VLDL Konzentration im Serum der Versuchstiere gefunden.
In der vorliegenden Arbeit sollte die Frage untersucht werden, ob die Überexpression der Lysosomalen Sauren Lipase in diesen Versuchtieren zu einer Veränderung der Aufnahmekinetik von Cho-lesterolestern in die Zellen führt. Hierdurch sollte ein Erklärungsansatz für die bei der Charakterisie-rung der Mauslinie gefundenen Veränderungen im Lipidstoffwechsel erbracht werden .Die intrazellulär erhöhte Aktivität der Lysosomalen Sauren Lipase führt zu einer vermehrten Hydrolase von Choleste-rolestern zu freiem Cholesterol und freien Fettsäuren. Hierbei könnte durch den höheren Substrat-durchsatz in futile cycle eine gesteigerte Aufnahme aus dem Blut in die Zellen ermöglicht werden, so dass es über die höhere Aufnahme von Lipiden zu der vermehrten Einspeicherung kommt. Um diese Frage zu beantworten, sollten Turnoverexperimente durchgeführt werden, in denen die Kinetik der Aufnahme von markierten Cholesterolestern gemessen werden kann

Higher adenoma detection rates with endocuff-assisted colonoscopy - a randomized controlled multicenter trial.
The Endocuff is a device mounted on the tip of the colonoscope to help flatten the colonic folds during withdrawal. This study aimed to compare the adenoma detection rates between Endocuff-assisted (EC) colonoscopy and standard colonoscopy (SC).This randomized prospective multicenter trial was conducted at four academic endoscopy units in Germany.500 patients (235 males, median age 64[IQR 54-73]) for colon adenoma detection purposes were included in the study. All patients were either allocated to EC or SC. The primary outcome measure was the determination of the adenoma detection rates (ADR).The ADR significantly increased with the use of the Endocuff compared to standard colonoscopy (35.4%[95% confidence interval{CI} 29-41%] vs. 20.7%[95%CI 15-26%], p<0.0001). Significantly more sessile polyps were detected by EC. Overall procedure time and withdrawal time did not differ. Caecal and ileum intubation rates were similar. No major adverse events occurred in both groups. In multivariate analysis, age (odds ratio [OR] 1.03; 95%[CI] 1.01-1.05), male sex (OR 1.74; 95%CI 1.10-2.73), withdrawal time (OR 1.16; 95%CI 1.05-1.30), procedure time (OR 1.07; 95%CI 1.04-1.10), colon cleanliness (OR 0.60; 95%CI 0.39-0.94) and use of Endocuff (OR 2.09; 95%CI 1.34-3.27) were independent predictors of adenoma detection rates.EC increases the adenoma detection rate by 14.7%(95%CI 6.9-22.5%). EC is safe, effective, easy to handle and might reduce colorectal interval carcinomas.ClinicalTrials.gov NCT02034929

funding programmes and valid questions concerning the relationship between
identity politics, knowledge and power - have applauded the formulation of
and research into various forms of Indigenous Knowledge Systems (lKS).
This interest in and advocacy for IKS, at times formulated in the context of
debates on postcolonial identity as an alternative to paradigms constructed on
particular Western-scientific assumptions, has run parallel to a more general
shift in thinking about development initiatives as requiring participatory
research methodologies and bottom-up implementation strategies. Within
development studies and anthropology, this shift has stimulated much
research on localised knowledge practices. There is, however, little evidence
that this body of anthropological (and ethnographic) literature has informed
the thinking of those writing and working within the theoretical paradigm of
Indigenous Knowledge Systems (lKS). Making use of a number of recent
ethnographic studies on Africa, the author argues for a theoretical
reconsideration of the IKS paradigm. He highlights important criticisms of the
ways in which many indigenous knowledge systems proponents essentialise
concepts such as knowledge and culture, as well as the methodological
limitations of much current IKS research. It is argued that a focus on the nonverbal
and local knowledge embodied in everyday practices, as well as the
performance of such knowledge, signals not only the limitations of much IKS
research but also redirects our attention to reformulating and invigorating
ideas about much needed local research

‘We are running for a living’ : work, leisure and speculative accumulation in an underground numbers lottery in Johannesburg
In this paper the author presents a historically-informed ethnography of a Johannesburg underground lottery. The meaning of this lottery is tied up with the local-level sociological organisation of lottery banks and the various actors who participate in it, with changing notions of social class, work and leisure under the conditions of growing inequality and jobless economic growth, and with the everyday strategies and agency of lottery runners and punters. The author uses the instance of this lottery to argue for a contextualised, multi-leveled and historically-grounded interpretation of the notions 'occult economies' and 'mysterious modes of accumulation' (Comaroff & Comaroff 1999a, 2000). The prominence of speculative accumulation in the context of this lottery and in the livelihood strategies of those living at the margins of the state and society are strikingly similar to financial practices under the conditions of casino capitalism, financialisation and securitisation in financial markets.
