Determination of the Acute Oral Toxicity of the Bruchid Resistance Factor from Vigna radiate var. sublobata Acc. TC1966 on Male Sprague-Dawley Rats. Adviser: Dr. Desiree M. Hautea The crude extracts of VC1973A-R, VC1973A-S and TC 1966 were evaluated for acute toxicity. VC1973A-R and VC1973A-S represent a pair of near isogenic lines (NILs) of V. radiate for bruchid resistance factor while Acc. TC1966 (V. r. var sublobata) is the parent source of this factor. Only VC1973A-S does not harbor the bruchid resistance factor. Powdered seeds of VC1973A-R, VC1973A-S and TC1966 were extracted with phosphate buffered saline (PBS) and the resulting test material was administered to male Sprague-Dawley rats at a constant volume of 10 ml/kg body weight (bw). Single doses of various treatments (45 g/kg bw VC1973A-R, 22.5 g/kg bw VC1973A-R, 20 g/kg bw TC1966, 10 g/kg bw TC1966, 45 g/kg bw VC1973A-S and 0 g/kg bw or pure PBS) were given to the animals using the gavage technique. The crude extracts of VC1973A-R, VC1973A-S and TC 1966 did not exhibit lethality even at high dose levels. However, 10 g/kg bw TC1966 crude extracts slowed down the recovery of growth of treated rats but not throughout the 14-day period. Enlarged spleen was observed in VC1973A-S treated rats. Histological evaluation revealed that 20 g/kg bw TC1966 caused severe disintegration of the mucosa. Based on these results, it was concluded that the presence of bruchid resistance factor in VC1973A-R does not confer any form of acute toxicity towards male Sprague-Dawley rats
