    An RNA interference knock-down of nitrate reductase enhances lipid biosynthesis in the diatom Phaeodactylum tricornutum

    When diatoms are stressed for inorganic nitrogen they remodel their intermediate metabolism and redirect carbon towards lipid biosynthesis. However, this response comes at a significant cost reflected in decreased photosynthetic energy conversion efficiency and growth. Here we explore a molecular genetics approach to restrict the assimilation of inorganic nitrogen by knocking down nitrate reductase (NR). The transformant strain, NR21, exhibited about 50% lower expression and activity of the enzyme but simultaneously accumulated over 40% more fatty acids. However, in contrast to nitrogen-stressed wild-type (WT) cells, which grow at about 20% of the rate of nitrogen-replete cells, growth of NR21 was only reduced by about 30%. Biophysical analyses revealed that the photosynthetic energy conversion efficiency of photosystem II was unaffected in NR21; nevertheless, the plastoquinone pool was reduced by 50% at the optimal growth irradiance while in the WT it was over 90% oxidized. Further analyses reveal a 12-fold increase in the glutamate/glutamine ratio and an increase NADPH and malonyl-CoA pool size. Transcriptomic analyses indicate that the knock down resulted in changes in the expression of genes for lipid biosynthesis, as well as the expression of specific transcription factors. Based on these observations, we hypothesize that the allocation of carbon and reductants in diatoms is controlled by a feedback mechanism between intermediate metabolites, the redox state of the plastid and the expression and binding of transcription factors related to stress responses.Peer reviewe

    Mirror Symmetry in Dimension 1 and Fourier–Mukai Transforms

    In this paper we will describe an approach to mirror symmetry for appropriate one-dimensional DM stacks of arithmetic genus g ≤ 1, called tcnc curves, which was developed by the author with Treumann and Zaslow in Sibilla et al. (Ribbon Graphs and Mirror Symmetry I, arXiv:1103.2462). This involves introducing a conjectural sheaf-theoretic model for the Fukaya category of punctured Riemann surfaces. As an application, we will investigate derived equivalences of tcnc curves, and generalize classic results of Mukai on dual abelian varieties (Mukai, Nagoya Math. J. 81, 153–175, 1981)

    Approximation and interpolation by large entire cross-sections of second category sets in Rn+1

    In [M.R. Burke, Large entire cross-sections of second category sets in Rn+1Rn+1, Topology Appl. 154 (2007) 215–240], a model was constructed in which for any everywhere second category set A⊆Rn+1A⊆Rn+1 there is an entire function f:Rn→Rf:Rn→R which cuts a large section through A in the sense that {x∈Rn:(x,f(x))∈A}{x∈Rn:(x,f(x))∈A} is everywhere second category in RnRn. Moreover, the function f can be taken so that its derivatives uniformly approximate those of a given CNCN function g in the sense of a theorem of Hoischen. In the theory of the approximation of CNCN functions by entire functions, it is often possible to insist that the entire function interpolates the restriction of the CNCN function to a closed discrete set. In the present paper, we show how to incorporate a closed discrete interpolation set into the above mentioned theorem. When the set being sectioned is sufficiently definable, an absoluteness argument yields a strengthening of the Hoischen theorem in ZFC. We get in particular the following: Suppose g:Rn→Rg:Rn→R is a CNCN function, ε:Rn→Rε:Rn→R is a positive continuous function, T⊆RnT⊆Rn is a closed discrete set, and G⊆Rn+1G⊆Rn+1 is a dense GδGδ set.     A comparison of Landau-Ginzburg models for odd-dimensional Quadrics

    In [Rie08], the second author defined a Landau-Ginzburg model for homogeneous spaces G/P, as a regular function on an affine subvariety of the Langlands dual group. In this paper, we reformulate this LG model (X^, W_t) in the case of the odd-dimensional quadric, as a rational function on a Langlands dual projective space, in the spirit of work by R. Marsh and the second author for type A Grassmannians and by both authors for Lagrangian Grassmannians. We also compare this LG model with the one obtained independently by Gorbounov and Smirnov, and we use this comparison to deduce part of a conjecture of the second author for odd-dimensional quadrics

    Liftings and the property of Baire in locally compact groups

    “Distant death” in Maxim Gorky's short story “Obsession”

    The paper analyzes the short story “Obsession” written by Maxim Gorky in the Nizhny Novgorod period of his work, which has been given little attention in philological works. On the one hand, the author himself defined its genre as a Christmas tale; on the other hand, this work cannot be brought into line with Christmas tales and short novels by Gogol and Dostoyevsky, since in Gorky’s story, no miracle occurs. However, this small text still deserves literary scholars’ attention. The short story introduces an interesting paradox of artistic space and time: in outward appearance, the action takes place within one room, on the couch, but the hero’s internal experiences, his conflict with the alter ego carry the reader into the distant past, the Christmas days of the main character’s family, and then the imagination, vision that visited Foma Mironovich come to the fore and become a plot-forming feature. The form in which the story content is presented (obsession, dream, delusion) is typologically similar to the structure of Russian folklore tales telling about encountering the phenomena of the “other world”. The results of the study may be of interest to both literary and cultural scholars

    In 2014, in special circumstances, the Volos Theological Academy in Greece, published a book written by two authors about the relationships between the Orthodox Church and Communism. The first part of it (p. 11-259), signed by the English researcher Joachim G. Persoon, specialised in the history of the Ethiopian Church (he gained his PhD at the School of Oriental and African Studies from the University of London), is in fact the PhD thesis of the first author. Divided in six chapters, the book comprises a series of introductory elements (pp. 11-50) like the literature used by the author in the process of documentation (pp. 15-17), theoretical concepts (pp. 22-29, aesthetical aspects (pp. 30-39), and terminology (p. 39), a big exploration of the history of the monastic life from the area (pp. 51-117) and the most important aspects that defined this life during Darg’s Communist period. The author presents the main geographic aspects and shows how they influenced the development of the monastic life in the area, talks about the importance of the sacred dance for the Ethiopian religion and culture and about the importance of the rules in Ethiopian monasticism and about the importance of the abbeys in the process of evangelism, and presents some examples of important monasteries, chosen from the almost 1000 (p. 14) existing on that territory

    The productivity effects of decentralized reforms - an analysis of the Chinese industrial reforms

    The empirical literature on the effects of ownership has not distinguished between the effects of ownership and the effects of control. It has also generally ignored the dynamic effects of various ownership and control rights. Using a rich set of panel data about changes in China's state-owned enterprises, the author examines the static and dynamic effects of decentralizing ownership and control rights. He finds that productivity and growth rates improved significantly when reform improved the incentives for managers and employees to learn and to work hard - for example by decentralizing the rights to control wages, make production decisions, and appoint new managers. Increasing profit-retention rates and adopting performance contracts - conventionally viewed as the most important reforms for China's state enterprises - did not improve productivity much. Overall, decentralization accounted for a least 42 percent of productivity growth in Chinese state enterprises in the 1980s.
