VES induces an autophagic response in SGC-7901 cells.

A study on the inhibitory effect of matrine on gastric cancer SGC-7901 cells
The objective of this paper was to investigate the antitumour mechanism of action of matrine by studying its inhibitory effect on gastric cancer SGC-7901 cells. SGC-7901 cells were chosen, and cell-killing capacity of matrine on gastric cancer SGC-7901 cells was determined using MTT assay and single PI staining assay. The results showed that matrine had an inhibitory effect on gastric cancer SGC-7901 cells, which was somewhat dose-dependent. The study concluded that matrine has a significant in-vitro inhibitory effect on SGC-7901 tumour cells, influences cell cycle of SGC-7901 cells, and induces their apoptosis.Key words: Matrine; SGC-7901; MTT assa

<p>HDAC4 expression was determined in SGC-7901 cells transfected with empty vector (NC) or HDAC4 (<b>A</b>) and (<b>B</b>). HDAC4 expression was determined in SGC-7901 cells transfected with siRNA oligos targeting HDAC4 (si-HDAC4) or scrambled siRNA (si-NC) by qRT-PCR and western blot (<b>C</b>) and (<b>D</b>) (n = 4). Floating cells with stem cell properties in gastric cell line SGC-7901
Aims To obtain floating spheres from the adherent gastric cancer cell line SGC-7901 and to analyze the properties of the spheres. Methods Serum-free medium was applied to cultured SGC-7901 cells. Limiting dilution assay, tumor formation assay, microarray analysis, and real-time fluorescent quantitative PCR were used to test the stem cell properties of the spheres. Results A subpopulation of SGC-7901 cells formed floating spheres in serum-free medium. These cells showed enhanced tumorigenic ability and also highly expressed certain stem-cell-associated proteins. Conclusions We successfully propagated floating spheres with stem cell properties in the SGC-7901 gastric cell line. </jats:sec
Study of Esculetin-Induced Apoptosis in Human Gastric Carcinoma SGC-7901 Cells
In order to investigate the effect and its mechanism of esculetin on human gastric carcinoma SGC-7901 cells, the effect of esculetin on human gastric carcinoma SGC-7901 cells were investigated by MTT method in vitro studies. Then the morphological feature changes of apoptosis cells with esculetin of different concentration by transmission electron microscope were observed and the levels of Cty-c in SGC-7901 cells line using FCM method were analyzed. The results revealed that the Esculetin significantly inhibited the growth of human gastric carcinoma SGC-7901 cells and greatly decreased its growth rate with the esculetin concentration increased, and then the morphological feature changes of apoptosis cells had been observed. After 24h, esculetin could make that the concentration of Cty-c in endochylema and Caspase-3 were all increased. Thus the study showed that esculetin remarkably inhibited the growth of human gastric carcinoma SGC-7901 cells and induce its apoptosis.</jats:p
