    L-leucine partially rescues translational and developmental defects associated with zebrafish models of Cornelia de Lange syndrome

    Cohesinopathies are human genetic disorders that include Cornelia de Lange syndrome (CdLS) and Roberts syndrome (RBS) and are characterized by defects in limb and craniofacial development as well as mental retardation. The developmental phenotypes of CdLS and other cohesinopathies suggest that mutations in the structure and regulation of the cohesin complex during embryogenesis interfere with gene regulation. In a previous project, we showed that RBS was associated with highly fragmented nucleoli and defects in both ribosome biogenesis and protein translation L-leucine stimulation of the mTOR pathway partially rescued translation in human RBS cells and development in zebrafish models of RBS. In this study, we investigate protein translation in zebrafish models of CdLS. Our results show that phosphorylation of RPS6 as well as 4E-binding protein 1 (4EBP1) was reduced in nipbla/b, rad21 and smc3-morphant embryos, a pattern indicating reduced translation. Moreover, protein biosynthesis and rRNA production were decreased in the cohesin morphant embryo cells. L-leucine partly rescued protein synthesis and rRNA production in the cohesin morphants and partially restored phosphorylation of RPS6 and 4EBP1. Concomitantly, L-leucine treatment partially improved cohesinopathy embryo development including the formation of craniofacial cartilage. Interestingly, we observed that alpha-ketoisocaproate (alpha-KIC), which is a keto derivative of leucine, also partially rescued the development of rad21 and nipbla/b morphants by boosting mTOR-dependent translation. In summary, our results suggest that cohesinopathies are caused in part by defective protein synthesis, and stimulation of the mTOR pathway through L-leucine or its metabolite alpha-KIC can partially rescue development in zebrafish models for CdLS

    This dataset accompanies the article “The local environment influences salt tolerance differently in four Salicornia europaea L. inland populations” and contains measurements of biomass, antioxidant enzyme activities (peroxidase, catalase, superoxide dismutase), and stress biomarkers (malondialdehyde, hydrogen peroxide, proline) of shoots and roots, from four inland populations of S. europaea: Inowrocław and Ciechocinek (Poland), and Salzgraben Salzdahlum and Soltauquelle (Germany). Data are provided in spreadsheet format (.ods) with separate worksheets for each trait category, enabling assessment of population-specific physiological responses to salinity.</p

    Dans l'ombre de Le Corbusier: une approche de l' uvre domestique de Pierre Jeanneret

    [EN] What is known of Pierre Jeanneret's opera is mostly related to the work done in association with Le Corbusier. However, despite the amount of research on the figure of the modern Master, there is little attention on the importance of P. Jeanneret within the Atelier 35 rue de Sèvres. Even more enigmatic is the work of P. Jeanneret after the separation. Since housing was P. Jeanneret s main fascination as an architect, such work consisted in profound research and left insightful observations on housing and domestic space. This article wants to reflect on the themes and projects developed by Pierre Jeanneret, starting from his work in the Atelier 35 rue de Sèvres and from the prefabricated housing projects developed during the Second World War, moving through the period in Europe, with his reflection on the local character of the residence, and ending with his work in Chandigarh, the new Punjabi capital in India.[ES] Lo que se conoce de la obra de Pierre Jeanneret se relaciona, en su mayor parte, a los trabajos realizados en asociación con Le Corbusier que, a pesar de la cantidad de investigaciones sobre la figura del Maestro moderno, hay poca atención sobre la importancia de P. Jeanneret dentro del Atelier 35 rue de Sèvres. Mucho más enigmático es la obra de P. Jeanneret después de la separación, obra que dejo una profunda investigación sobre la vivienda y el espacio doméstico, puesto que la vivienda fue para P. Jeanneret su principal fascinación como arquitecto. Es así que en el presente artículo se quiere reflexionar sobre las temáticas y proyectos desarrollados por Pierre Jeanneret, desde su labor en el Atelier 35 rue de Sèvres, pasando por los proyectos de vivienda prefabricada desarrollados durante la segunda guerra y postguerra, una reflexión sobre el carácter local de la residencia para así finalmente llegar a su trabajo en Chandigarh, la nueva capital Punjabi en India.[FR] Ce qu on connaît de l opéra de Pierre Jeanneret est surtout lié au travail réalisé en association avec Le Corbusier. Cependant, malgré la quantité de recherches sur la figure du moderne Maître, on parle peu de l'importance de P. Jeanneret au sein de l'Atelier 35 rue de Sèvres. L' uvre de P. Jeanneret est encore plus énigmatique après la séparation. Vu que le logement était la principale fascination de P. Jeanneret en tant qu'architecte, un tel travail consistait en recherche donnant des observations perspicaces sur le logement et l'espace domestique. Cet article veut réfléchir sur les thèmes et projets développés par Pierre Jeanneret, à partir de son travail à l'Atelier 35 rue de Sèvres et des logements préfabriqués.  Ces projets ont été développés pendant la Seconde Guerre mondiale, traversant la période en Europe, avec une réflexion sur le caractère local de la résidence, et terminant par son travail dans Chandigarh, la nouvelle capitale du Pendjab en Inde.Cardenas Niño, O.
