TA CLONING PARTIAL ALBUMIN GENE AND ITS TEST FOR SUPPORTING PHARMACOGENOMIC DETECTION DEVELOPMENT
DNA recombinant technology is a tool that could support thedevelopment of pharmacogenomic detection such as for CYP2D6 copy number detection application. The housekeeping gene was identified as an internal control alternative for the copy number of the CYP2D6 detection component. There are many kinds of housekeeping genes, including albumin. In this report, we have successfully TA-cloned partial albumin to support the development of CYP2D6 copy number detection. TA-cloning was successfully confirmed with white-blue methods in media LB + ampicillin + IPTG + X-Gal and with colony PCR methods. Further, we made a standard curve with this recombinant plasmid to evaluate albumin qPCR efficiency and showed high-efficiency qPCR 0,9914 (99%). Based on the CYP2D6 copy number developed, the test in this study showed low concordance with high reproducibility Long PCR, indicating that the nucleotide base area of albuminand CYP2D6 in this studywas not recommended for CYP2D6 gene multiplication detection.
INVENTORY AND STUDY PHARMACOLOGICAL POTENTIAL OF PLANTS IN THE PAJANGAN HAMLET, WEDOMARTANI, NGEMPLAK, SLEMAN, YOGYAKARTA
This research aims to inventory and study pharmacological potention of plants in Pajangan Hamlet, Wedomartani, Ngemplak, Sleman, Yogyakarta as medicinal plants and live stalls. The research was conducted from June to October 2023 using observational methods. Plants were identified with PlantNet software and reference againts the Flora of Java book. In addition, the study of the utilization of these plants was carried out by literature study and distributing questionnaires to the residents. The results found 104 plant species with 19 different families can be identified in Pajangan Hamlet. The plants are categorized as herbs, ornamental plants/flowers, fruits, vegetables, and wild plants. These findings highlight the richness of biodiversity in Pajangan Hamlet with significant potentital for improving health and economic livelihoods through the use of local plant species in traditional practices and diets. Based on the result, it can be concluded that Pajangan Hamlet has a variety of plants that are widely used in traditional medicine and food, thereby underscoring the importance of local biodiversity in improving health and well-being
STUDENTS’ LEARNING ACTIVITIES IN PHYSICS CLASSES OF GRADE X BY IMPLEMENTING INQUIRY TRAINING MODEL
This research aims to analyze students’ learning activities in implementing the inquiry training learning model in grade X. The physics lesson material taken in this research is linear motion. SMAN Percut Sei Tuan is the location where the research was carried out. The samples in this study were two classes: grade X-9 which uses inquiry training model and X-5 which uses direct instruction model. The type of this research is quasi experiment. The observation sheet was used as an instrument in this research. The data in this research were analyzed using the independent sample t test to find out the differences in student learning activities using the inquiry training model and the direct instruction model. Student learning activities are observed during learning activities by an observer. The average of student learning activity in class X-9 is 80 and class X-5 is 71. Based on the results of data analysis, it was found that the sign value < 0,05. So it can be concluded that there are differences in student learning activities using the inquiry training model and the direct instruction model.
DEVELOPMENT OF AN RPG MAKER MV-BASED INTERACTIVE GAME AS LEARNING MEDIA ON VIRUS MATERIALS FOR GRADE X STUDENTS AT NURUL ISLAM JEMBER SENIOR HIGH SCHOOL
The limited accessibility of digital educational game resources on the topic of viruses in the public domain poses a significant challenge that must be addressed. The primary objective of this research is to determine the validity of the RPG maker MV based game, followed by an assessment of its practicality. The research used Lee & Owens as the development model. The result of this study is the produced RPG maker MV-based game, focused on virus material. The validity of the game has been confirmed through product validation conducted by experts in the discipline, achieving an average percentage of 96.63% (highly valid) from material experts, 93.07% (highly valid) from media experts, and 80.71% (valid) from biology teacher practitioners. The practicality of the RPG maker MV-based game was then demonstrated by small-scale trials, yielding an average performance rate of 93% (an exceedingly captivating product), and large-scale trials, yielding an average performance rate of 94.3% (an exceedingly captivating product)
ETHNOBOTANY SAGO (Metroxylon sago Rottb) in JAILOLO LOCAL COMMUNITY, WEST HALMAHERA REGENCY, INDONESIA
Sago plants have long played an important role in people's lives, particularly in eastern Indonesia—such as in Papua, Maluku, and North Maluku. The objective of this study was to investigate how the local community in Jailolo District, West Halmahera Regency, utilizes sago plants in line with their traditional wisdom. The research method employed is exploratory-descriptive, which is valuable for depicting the characteristics of a phenomenon. This research was conducted in Jailolo District, West Halmahera Regency, and lasted for 3 months, from January to February 2023. The methodology involved direct observation and collected information from Sahu ethnic by interviews with the snowball sampling method. Additionally, documentation of the various forms of sago plant utilization was undertaken, encompassing sago stems, bark, fronds, sago leaf stalks, sago leaves, and sago flour. The data collected in this study were analyzed using qualitative descriptive analysis and presented in a narrative format. The results revealed that a majority of the people in Jailolo District, West Halmahera Regency, utilized sago plants to fulfill their daily needs, using them as an alternative food source to substitute rice. Sago plants are utilized in various forms, primarily in the form of sago flour, which is used to create a range of processed foods such as bagea, sago macarons, popeda, boko-boko, sago plates, sago sugar, and sinyole. Additionally, sago leaves are utilized for roofing houses, while sago fronds are used for ceilings, walls, and crafts such as paludi (Saloi). Sago stems are used to extract starch (flour), while the bark is utilized as firewood
ANALYSIS OF THE DESIGN, IMPLEMENTATION, AND EVALUATION OF 21ST CENTURY SKILLS EDUCATION IN CLASS X AT STATE SENIOR HIGH SCHOOL
This research was conducted with the aim of describing 1). Analyzing the design of learning implementation, 2). Analyzing the learning process (RPP), 3). Analyzing the evaluation of 21st century learning in class X of SMAN 7 Bengkulu City. The research method used is a descriptive method, using qualitative descriptive analysis techniques. The samples used were students and physics teachers of class This research shows that the results of the analysis of learning implementation plans (RPP) are based on 21st century learning skills with a percentage of 86.87%, consisting of aspects, namely planning with a percentage of 87.50%, implementation with a percentage of 85.62%. and evaluation of 21st century learning with a percentage of 87.50%. So from the results of this analysis it can be concluded that the design for implementing 21st century skills learning is in the good category
DEVELOPMENT OF INTERACTIVE CONTENT IN WEBSITE LEARNING MEDIA TO ADDRESS STUDENT MISCONCEPTIONS ABOUT THE GREENHOUSE EFFECT
This study addresses the pressing need to improve students' comprehension of the greenhouse effect by leveraging innovative website-based Augmented Reality (AR) learning media. The purpose is to explore the effectiveness of this media in mitigating misconceptions surrounding the greenhouse effect while providing practical solutions for educators. The research methodology combines qualitative Design and Development (D&D) approaches with quantitative measures to comprehensively evaluate the impact of AR learning media on students' understanding. Results demonstrate a significant increase of over 50% in student comprehension post-media use, validated by expert assessments and positive teacher feedback. Students reported overcoming prior misconceptions through the media's interactive approach. The study emphasizes the necessity of identifying key elements for effective AR learning, recommending comprehensive teacher training, content updates, and long-term impact assessments. The holistic approach employed unveils insights into how learning media can effectively reduce misconceptions, thereby enhancing awareness of crucial environmental issues like the greenhouse effect
DEVELOPMENT OF SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS (STEM)-BASED E-BOOK ON HUMAN DIGESTIVE SYSTEM MATERIAL BASED ON SCIENTIFIC LITERACY
Abstract The research was conducted on students’ low scientific literacy skills in Indonesia based on PISA, PUSMENDIK data and the lack of science, technology, engineering, and mathematics (STEM)-based teaching materials that measure students’ scientific literacy. Researcher develop teaching material in the form of interactive E-book that can be used on all devices with feasible based on scientific literacy skills on human digestive system material. The research method used is research and development (R&D) with the analysis, design, development, implementation, and evaluation (ADDIE) from Branch. Subjects in this study was 4 experts (1 expert in media, learning design, material, and linguist). The instruments used are tests that experts and non-tests have validated. Test instruments assessment was given to 4 experts (1 expert in media, learning design, material, and linguist). The results showed that resulting validation from a media expert with x̄ = 98.3%, a material expert with x̄ = 83.3%, a learning design expert with x̄ = 95.3%, and a linguist expert with x̄ = 85%, and practically x̄ = 97, then an STEM-based E-book was declared feasible, and practically
THE EFFECT OF LIQUID ORGANIC FERTILIZER OF ALOE VERA (Aloe vera L.) AND COFFEE GROUNDS ON THE GROWTH AND YIELD OF CAYENNE PEPPER PLANTS (Capsicum frutescens L.)
This research aims to determine the effect of liquid organic fertilizer from aloe vera (Aloe vera L.) and coffee grounds on the growth and yield of cayenne pepper plants (Capsicum frutescens L.). The type of research used by researchers is experimental research with a Randomized Group Factorial Design (RGFD). The samples used were 75 cayenne pepper (Capsicum frutescens L.) seeds of the Bara variety. The observation parameters used in this research were plant height (cm), number of leaves (strands), and plant fresh weight (grams). The results of data analysis showed that there was no effect of liquid organic fertiIizer from aloe vera (Aloe vera L.) and coffee grounds on the growth and yield of cayenne pepper plants (Capsicum frutescens L.). In the plant height parameter is smaller than , namely 1,070 < 1,85 at the age of 14 DAP, 0,978 < 1,85 at the age of 28 DAP, 1,747 < 1,85 at the age of 42 DAP, 1,373 < 1,85 at the age of 56 DAP, 1,238 < 1,85 at age 70 DAP, and 1,096 < 1,85 at age 84 DAP. In the number of leaves parameter is smaller than 0,738 < 1,85 at the age of 14 DAP, 1,059 < 1,85 at the age of 28 DAP, 0,981 < 1,85 at the age of 42 DAP, 0,833 < 1,85 at the age of 56 DAP, 0,154 < 1,85 at age 70 DAP, and 0,798 < 1,85 at age 84 DAP. In the fresh fruit weight parameter, is smaller than for 84 DAP, namely 0,798 < 1,85
DEVELOPMENT OF FLIPPING BOOK LEARNING MATERIALS INTEGRATED WITH PROBLEM-BASED LEARNING TO IMPROVE STUDENTS' DIGITAL LITERACY
Digital literacy is the ability to access, manage, understand, evaluate and create information safely and appropriately. This research aims to produce a valid, practical and effective problem-based learning flipping book instructional material focusing on the diversity of mangrove species, Gonda Mangrove Park, Polewali Mandar to increase student’s digital literacy. This research is R&D and follow the ADDIE instructional design model. The population in this study were X4 SMAN 2 Majene, Sulawesi Barat. The research instrument used pretest-posttest, questionnaires and digital literacy observation sheets. Data analysis through N-Gain Test. The findings obtained from this study indicate a percentage of validity test such as material 100%, learning instruction 81.42%, lesson plan 95.75% and educational practitioner 100%. Practicality test including one-to one trial 83.17%, small group trial 86.90% and field trial 88%, three of them categorized as practical. Based on the N-Gain score, the effectivity of instructional materials shows 0.71, it means the learning materials increasing student’s digital literacy effectively
