The dataset generated and analyzed during the current study was collected at the University of Debrecen, Faculty of Medicine, in May 2024. It comprises anonymized questionnaire responses and simulator performance data from 82 medical students (1st–6th year) who participated voluntarily in the da Vinci Skills Simulator (dVSS) assessment. The dataset includes demographic and background variables (e.g., academic year, gender, handedness, and hobbies requiring manual dexterity) as well as machine-recorded performance indicators (overall score, efficiency, and penalty scores) obtained during the “Sea Spikes 1” (EndoWrist) task. Due to ethical and privacy considerations, the dataset is not publicly available; however, de-identified data may be made available from the corresponding author upon reasonable request and with permission from the Regional and Institutional Research Ethics Committee of the Clinical Centre of the University of Debrecen (approval number: DE RKEB/IKEB: 6775-2024)
Cricetus cricetus bioinformatic data processing
This is the bioinformatic pipeline we run to produce filtered vcf files for downstream analyses from raw reads. The steps include: (1.) demultiplexing, (2.) quality and sequence filtering, (3.) mapping to the reference, (4.) variant calling, (5.) basic filtering of vcf (imiss, meanDP, maf, biallelic, lmiss), (6.) LD pruning and keeping only SNPs. The vcf produced after step (5.) is suitable for phylogenetic tree reconstruction, the vcf produced after the last step is suitable for population genomics
Interplay between dysregulated immune system and the footprints of blood-borne miRNAs in treatment naive Crohn's disease and ulcerative colitis patients
Background: Dysregulated T cell-mediated immune response is the hallmark of inflammatory bowel diseases (IBD): Crohn’s disease (CD) and ulcerative colitis (UC). miRNAs are established regulators of biological processes, with clear contribution to the pathophysiology of several diseases. Here, we attempt to elucidate the correlation between dysregulated immune system and altered miRNA signatures in IBD patients. Furthermore, we also aim to identify differentially expressed miRNAs that could potentially serve as differential diagnostic markers of CD and UC.
Methods: High-throughput small RNA sequencing was conducted to quantify circulating miRNAs in 27 participants (8 CD, 10 UC and 9 healthy volunteers). To describe immune-dysregulation, circulating T-cell related cytokines were determined via bead array. Additionally, in IBD patients, the distribution of T cell subpopulations was also determined in circulation and in tissue biopsy samples using multicolor flow cytometry.
Results: Based on small RNA sequencing results, 14 miRNAs showed significant expression differences between IBD and control group after qRT-PCR validation. Five miRNAs were found to exhibit significantly different expression and high discriminatory potential (AUC:0.7-1) between CD and UC patients. Furthermore, circulating IL-22 and IFN-γ levels were significantly higher in IBD. Notably, miRNAs exhibited several strong negative and positive correlations with cytokine levels and with T-cell subset distribution in circulation and tissue samples, with disease specific association pattern.
Conclusion: We identified differentially expressed miRNAs in IBD patients’ groups. A subset of these miRNAs shows promising diagnostic value for distinguishing CD from UC. Thereby scrutinizing miRNA expression in blood samples of IBD patients might holds diagnostic and monitoring potential. Our results confirmed that, dysregulated miRNAs reflect the underlying immune dysregulation and may contribute to IBD diseases pathology
